C=C double bond reduction kit
The enzymes in this kit (EZK002) have been selected for their activity on a wide selection of substrates and in a wide range of pH, to encounter the different synthetic needs of our customers.
This kit contains 7 enzymes for the reduction of C=C double bonds in presence of an electron withdrawing group (EWG). Examples of electron withdrawing groups are carbonyls (ketones and aldehydes) and nitro. In some specific instances, carboxylic acids, esters and nitriles can also act as activating groups.
The ENEs in this kit include prokaryotic and eukaryotic wild-type enzymes. Most ENEs contain a flavin cofactor (FMN or FAD) that is incorporated in the enzyme at the expression stage and is fundamental for catalysis. Recently a class of ENEs have been characterised that do not require flavin cofactor. The enzyme ENE-105 included in this kit is a member of this class.

*ENE-105 is also capable to convert substrates with an ester as the electron withdrawing group
All ENEs require an additional cofactor (NADH or NADPH) for reducing the C=C double bond. To be able to use this cofactor in catalytic amounts, the oxidised cofactor has to be continuously reduced, this is called cofactor regeneration. The enzymes accept either or both NAD(P)H cofactors and their regeneration can be achieved by adding a cofactor regeneration enzyme (GDH or FDH, also provided in the kit) and its substrate (D-glucose or sodium formate, respectively).
We supply each kit with 200mg of each enzyme, except GDH-101, which is 500mg and also comes with a supply of relevant cofactors.
